Each school has an opposite school except Balance, and then tend to do more damage against their opposites.
Fire -Ice, Storm - Myth, Life - Death.
As far as questing, each opponent or boss that you fight will have a bonus when using attacks from it's own school, and also a weakness against damage recieved from the opposite school.
There are exceptions of course and sometimes you will do extra damage against Balance enemies or Sun\Solar opponents.

The schools which are opposites:

Life -- Death
Storm -- Myth
Fire -- Ice

Balance has no opposite school.

Fire beats Life, and Ice

Storm beats Fire, and Myth

Balance loses to the three spirit classes.

Fire and Ice
Life and Death
Myth and Storm

Balance has no opposite but Death and Myth seem to have a slight boost.
